2. Objective : 1. Finding the diversity of freshwater Protist
2. Drawing the Protist found
3. Describing the characteristic of protest based on observation
4. Identifying Protist based on the morphological characteristic
5. Classifying Protist based on their taxonomical position
3. Tools and Materials
- Microscope 1 piece/group
- Object glass 4 pieces
- Cover glass 4 pieces
- Bottle sampler 4 pieces
- Pipette 1 piece
- Sewage water
- Soaking of dried rice stalk
- Water from the pool
- Water the well
- HCl 2 % or Formalin 4 %
4. Procedure
- Labeled bottle with A for sewage water, bottle B for soaking of dried stalk, bottle C for water from the pool, and bottle D for water from the well
- Prepare the microscope and set up to be ready for used
- Take the water from bottle A by pipette, drop the small one to the object glass and covered by the cover glass
- Observe under the microscope by 5x10 magnification
- If the organism observed is too active moving, drop it with HCL 2% or Formalin 4%
- Increase the magnification by 10x10 or 40x10, and draw the organism found in the observation table
- Repeat the same procedure with the sample from bottle B, C and D
- Discuss the result and answer some questions in the end of this manual work
